FOXO4-DRI (FOXO4 D-Retro-Inverso Peptide)

FOXO4-DRI, or FOXO4 D-Retro-Inverso Peptide, is an experimental peptide investigated in cellular senescence and senescent-cell biology research.

FOXO4 is a member of the Forkhead Box O family of transcription factors and is associated with multiple biological processes, including cellular stress responses, DNA-damage responses, cell-cycle regulation, and cell survival.

A major area of FOXO4-DRI research involves the interaction between FOXO4 and the p53 protein. In certain preclinical models, researchers have investigated how disrupting the FOXO4–p53 interaction may influence senescent-cell survival and apoptosis-related signaling.

FOXO4-DRI therefore provides an experimental tool for studying mechanisms involving cellular senescence, senescent-cell selectivity, p53 signaling, apoptosis, and tissue aging.

The peptide has also attracted interest in senolytic research, where investigators study relationships between the accumulation of senescent cells, age-associated tissue changes, and experimental strategies targeting senescent-cell biology.

Research involving FOXO4-DRI remains primarily experimental and preclinical. Current evidence does not establish that FOXO4-DRI safely or effectively eliminates senescent cells in humans, reverses human aging, extends lifespan, or treats age-related diseases.
